Transcriptional profiling of Pseudomonas aeruginosa ∆lasR in mono- and co-culture with wild type.

(Submitter supplied) Pseudomonas aeruginosa is a troublesome opportunistic pathogen isolated from diverse environmental sources. An arsenal of degrading enzymes and antagonistic factors contribute to P. aeruginosa persistence and damage of a susceptible host. Largely through density-dependent regulation referred to as quorum sensing, the LasR, RhlR, and PqsR transcription factors collectively modulate hundreds of genes, including the expression of several virulence factors, in response to diffusible signals called autoinducers. more...

Genetic and transcriptomic characteristics of RhlR-dependent quorum sensing in cystic fibrosis isolates of Pseudomonas aeruginosa: isolate E167

(Submitter supplied) In people with the genetic disease cystic fibrosis (CF), bacterial infections involving the opportunistic pathogen Pseudomonas aeruginosa are a significant cause of morbidity and mortality. P. aeruginosa uses a cell-cell signaling mechanism called quorum sensing (QS) to regulate many virulence functions. One type of QS consists of acyl-homoserine lactone (AHL) signals produced by LuxI-type signal synthases, which bind a cognate LuxR-type transcription factor. more...

Using RNA Seq to define the regulon of the Pseudomonas aeruginosa transcription factor Anr in low-oxygen colony biofilms.

(Submitter supplied) Purpose : The goal of this study was to use RNA Seq to define the regulon of the transciption factor Anr by comparing global transcriptional profiles of Pseudomonas aeruginosa strain PAO1 and a clinical isolate with their isogenic ∆anr mutants, grown in colony biofilms at 1% oxygen. Methods : mRNA profiles were generated for laboratory strain PAO1 and for a clinical isolate J215, as well as for ∆anr derivatives of each strain, in duplicate, by deep sequencing. more...

PqsE expands and differentially modulates the RhlR quorum sensing regulon in Pseudomonas aeruginosa

(Submitter supplied) While it is known that PqsE can stimulate the ability of RhlR to control some virulence factors, no data are available allowing clear discrimination of the PqsE and RhlR regulons. In this study, a P. aeruginosa PAO1 mutant strain with deletion of multiple QS elements and inducible expression of PqsE and/or RhlR has been generated and validated. RNA-seq performed in this genetic background allowed to unambiguously define the regulons controlled by PqsE and RhlR when produced alone or in combination Data produced in this study demonstrate that PqsE mainly impacts on the P. more...

Lyngbyoic Acid, a "Tagged" Fatty Acid from a Marine Cyanobacterium, Disrupts Quorum Sensing in Pseudomonas aeruginosa

(Submitter supplied) Quorum sensing (QS) is a mechanism of bacterial gene regulation in response to increases in population density. Production of small molecule QS signals, their accumulation within a diffusion-limited environment and their binding to the LuxR-type receptor trigger QS-controlled gene regulatory cascades. QS pathways mediated by acylhomoserine lactones (AHLs) in Gram-negative bacteria are the best studied. more...

Analysis of the organization and expression patterns of the convergent lasR/rsaL gene pair uncovers mutual influence

(Submitter supplied) The two adjacent genes encoding the major quorum sensing regulator, LasR, and its opponent, RsaL, overlap in their coding 3´ends and produce mRNA transcripts with long untranslated 3´ends that overlap with the sense transcripts of the gene on the opposing DNA strand. In this study, we evaluated whether the overlapping genes are involved in mutual regulatory events, such as transcriptional interference and interference by natural antisense transcripts. more...

A Distinct QscR Regulon in the Pseudomonas aeruginosa Quorum Sensing Circuit

(Submitter supplied) To better understand the role of QscR in P. aeruginosa gene regulation and to better understand the relationship between QscR, LasR and RhlR control of gene expression we used transcription profiling to identify a QscR-dependent regulon. Our analysis revealed that QscR activates some genes and represses others. Some of the repressed genes are not regulated by the LasR-I or RhlR-I systems while others are. more...

Pseudomonas aeruginosa PA14, mvfR and anthranilic acid analogs

(Submitter supplied) The Pseudomonas aeruginosa MvfR-dependent QS regulatory pathway controls the expression of key virulence genes; and is activated via the extracellular signals 4-hydroxy-2-heptylquinoline (HHQ) and 3,4-dihydroxy-2-heptylquinoline (PQS), whose syntheses depend on anthranilic acid (AA), the primary precursor of 4-hydroxy-2-alkylquinolines (HAQs). We identified halogenated AA analogs that specifically inhibited HAQ biosynthesis and disrupted MvfR-dependent gene expression. more...

Expression profile for PA14 lasR rhlR double mutant with H2O2 stress

(Submitter supplied) Quorum sensing controls the expression of multiple virulence factors. PA14 genes lasR and rhlR are necessary for quorum sensing via homoserine lactones. A PA14 lasR rhlR deficient mutant exhibits a reduced oxidative stress response. Here we conducted a microarray to determine oxidative stress response gene regulation mediated by the homoserine lactone quorum sensing circuits.

Pseudomonas aeruginosa PA14, pqsE and PQS

(Submitter supplied) The Pseudomonas aeruginosa MvfR-dependent QS regulatory pathway controls the expression of key virulence genes; and is activated via the extracellular signals 4-hydroxy-2-heptylquinoline (HHQ) and 3,4-dihydroxy-2-heptylquinoline (PQS). Our findings reveal a multi-layered homeostatic regulation of PQS and HHQ and the MvfR regulon component PqsE which is a key mediator in orchestrating this homeostatic regulation.

Regulation by growth temperature of Pseudomonas aeruginosa quorum-sensing depedent virulence factors production involves two RNA-thermometers.

(Submitter supplied) Several bacterial human pathogens regulate the production of virulence factors by temperature, expressing them only at 37 °C. Accordingly we show that the production of all P. aeruginosa virulence factors that are dependent on the QS transcriptional regulator RhlR, but only a fraction that are activated by LasR, are induced at 37 °C compared to 30 °C or 25 °C. The RhlR-dependent induction at 37 °C is a posttranscriptional effect due to an RNA thermometer of the ROSE family that thermoregulates the expression of rhlAB operon involved in rhamnolipids production, a virulence associated trait. more...

Pseudomonas virulence factor controls expression of virulence genes in Pseudomonas entomophila

(Submitter supplied) Quorum sensing is a communication strategy that bacteria use to collectively alter gene expression in response to cell density. Pathogens use quorum sensing systems to control activities vital to infection, such as the production of virulence factors and biofilm formation. The Pseudomonas virulence factor (pvf) gene cluster encodes a quorum sensing system (Pvf) that is present in over 500 strains of proteobacteria, including strains that infect a variety of plant and human hosts. more...
