SRX541304: Lysogeny Broth Transcriptome of HM27
1 ILLUMINA (Illumina HiSeq 2000) run: 44.3M spots, 7G bases, 3.4Gb downloads

Design: E. coli (HM27) isolated from urine sample of a patient with urinary tract infection, was cultured in lysogeny broth (1% tryptone, 0.5% yeast extract, 0.05% NaCl) to mid-exponential phase, statically at 37°C. Cultures were stabilized by the addition of RNA-protect reagent (Qiagen). RNA was isolated with RNAeasy mini kit (Qiagen). Contaminating DNA was removed by treatment with TurboDNAase (Life Technologies). RNA was treated with MICROBEnrich Kit (Life Technologies), so that this sample can be directly compared with urinary tract infection RNA samples. The RNA was reverse transcribed to cDNA and the next generation sequencing libraries were generated using the Ovation Prokaryotic RNA-Seq system (NuGen) and the Encore next generation sequencing (NGS) library system (NuGen). The libraries were sequenced using an Illumina HiSeq2000 by the Genome Resource Center at the Institute for Genome Sciences, University of Maryland, Baltimore.
