Design: RIT714 Was isolated by placing a wild mushroom fruiting body in LB broth. Genomic DNA from single colonies, plated onto the corresponding agar plates, was isolated using the QIAGEN DNeasy UltraClean Microbial Kit. Genomic DNA was quantified using a Qubit 3.0 fluorometer, and the genomic DNA was processed using the Nextera XT (Illumina) library preparation kit for sequencing using the MiSeq

The purpose of this study was to identify endophytic bacteria from wild mushrooms in a park near Rochester, NY, USA. Wild mushrooms were homogenized, and bacteria from the homogenate were isolated on agar plates. Single colonies were subcultured, and genomic DNA from single colonies was extracted and subjected to library preparation and whole-genome sequencing on an Illumina MiSeq.
