SRX11703087: Leclercia sp. EMC7 genome sequencing
1 ION_TORRENT (Ion Torrent S5) run: 11.5M spots, 2.3G bases, 1.7Gb downloads

Design: Genomic DNA was isolated from LB-grown stationary-phase cells using PureLink genomic DNA isolation kit (Thermo Fisher Scientific, USA)following manufacturer's protocol. DNA was quantified using a SPECTROstar Nano microplate reader (BMG Labtech, Germany). DNA shearing and genomic DNA library preparation was done using an Ion Xpress Plus fragment library kit (Thermo Fisher Scientific, USA) and the library was sequenced on a 530 chip of the Ion S5 system (Thermo Fisher Scientific). Before sequence retrieval from Ion S5, low-quality and polyclonal reads were filtered out and the adaptor sequences trimmed using the inbuilt PGM software. Then adapter-free, quality-filtered reads were obtained and subsequently assembled into contigs using SPAdes v3.13.0 with default parameters.

show Abstracthide Abstract
This project aims to discover gut-microbes of earthworm (Eisenia fetida), that are capable of metabolizing lethal nitrotoxin 3NPA. This project also aims to reveal genome sequences of such microbes employing NGS technologies. This may help us understand how this annelid survives in environments contaminated with such toxins and the benefits it derives from the gut inhabitants in tackling such stress conditions.
