1 ILLUMINA (Illumina NovaSeq 6000) run: 25.2M spots, 5.1G bases, 1.5Gb downloads

Design: The ATCC14917 cells were grown in the acidified broth at 37C until they reached an OD600 of 0.40.5. Then the cells were cultured in the presence of 40 mM NaNO2 and incubated for 1 h. An RNeasy Protect Bacteria Mini Kit was used to extract the RNA from the pellets.RNA sequencing was performed by Macrogen, Inc. The NEBNext rRNA Depletion kit was used to remove ribosomal RNAs. Libraries for Illumina se- quencing were prepared using a TruSeq Stranded mRNA Sample Prep Kit. We used an Illumina NovaSeq (Illumina) machine to sequence the libraries using paired-end sequencing (2 100 bp).

Study: genome sequencing of Lactiplantibacillus plantarum LP7 and its transcriptome profiles under nitrite stress

We isolated bacteria resistant to nitrite stress from human feces and performed whole-genome sequencing to identify them as Lactiplantibacillus plantarum LP7 (LP7). To identify the gene related to nitrite stress in LP7, we performed RNA-seq of LP7 with L. plantarum ATCC14917 (ATCC14917) as a reference strain. Transcriptome profiles were obtained using illumina NovaSeq. RNA samples were prepared in biologically independent duplicates with/without nitrite in LP7 and in ATCC14917.
