SRX4636333: Draft genome sequence of Lactobacillus fermentumLf2, an exopolysaccharide-producing strain isolated from Argentine cheese

Design: Genome of L. fermentum Lf2 was determined by GenProbio srl (Parma, Italy) using the MiSeq Illumina (Illumina, USA). Genomic libraries were constructed employing the TruSeq DNA PCR-Free LT Kit (Illumina) and using 2.5g of genomic DNA, which was fragmented with a Bioruptor NGS ultrasonicator (Diagenode, USA) followed by size evaluation using Tape Station 2200 (Agilent Technologies). Library samples were loaded into a Flow Cell V3 600 cycles (Illumina) according to the technical support guide, and generated reads were depleted of adapter sequences, quality filtered and assembled through the MEGAnnotator pipeline (Lugli et al, 2016, FEMS Microbial Lett).

Lactobacillus fermentum LF2 was isolated from regional Tybo cheese. It's most interesting characteristic is the ability to produce high concentrations (~1g/L of crude extract) of exopolysaccharide (EPS). The EPS extract displayed the property of improving the texture and rheology of yogurt as well as protecting mice from Salmonella infection when added to milk or yogurt. Preliminary chemical characterization shows that the EPS is mainly composed of beta-glucan (a homopolysaccharide) and a heteropolysaccharide with glucose and galactose in its structure. The LF2 draft genome sequence harbours three gene clusters potentially involved in EPS production.
